2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,磺胺嘧啶,磺胺甲基嘧啶,磺胺-5-甲氧嘧啶,磺胺甲恶唑,磺胺间二甲氧嘧啶标准品购于xxx
All chemicals and solvent were HPLC. Standard: sulfadiazine, sulfamerazine, sulfamethoxydiazine, sulfamethoxazole, sulfadimethoxypyrimidine and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取2.0g左右的样品;
2) 2.0g样品加入5 mL甲醇与xxx缓冲液等体积混合溶液,振荡10 min,超声提取10 min,离心;
3) 收集上清液,残渣按上述方法重复提取2次,合并提取液,浓缩至5 mL;
4) 浓缩液通过LC-SAX与LC-18串联柱,萃取富集;
5) 用6 mL高纯水清洗串联柱,真空干燥10 min;
6) 拆下LC-SAX小柱,用3 mL甲醇洗脱LC-18小柱,收集洗脱液,浓缩至干;
7) xxx用甲醇-水(60 : 40)定容至1 mL,摇匀并经0.22 µm滤膜过滤,进样20 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Waters高效液相色谱仪,配备紫外检测器。
色谱柱为xxx-C18 (150 mm*4.6 mm, 5 μm),流动相为乙腈-0.01 mol/L xxx(20:80, V/V),流速为1.0 mL/min。
