2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级，亚麻酸，亚油酸，棕榈酸，油酸，硬脂酸标准品购于xxx
All chemicals and solvent were HPLC. Standard: linolenic acid, linoleic acid, palmitic acid , oleic acid, stearic acid and were purchased from xxx.

2.2 样品处理Sample Preparation
1) 将样品摇匀，称取1.0 g左右的样品；
2) 1.0 g样品中加入50 mL 3 mol/L xxx-乙醇溶液，60℃水浴皂化20 min；
3) 冷却，加入20 mL蒸馏水，混匀，用盐酸调pH至3.0～4.0；
4) 用无水乙醚萃取2次，合并醚层；
5) 加入30 mL xxx反萃取乙醚相，醚层用氮气吹干；
6) 残留物用xxx定容至2 mL，并用0.22 µm滤膜过滤，进样15 µL。

2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Waters 2695型高效液相色谱仪，配备蒸发光散射检测器。
色谱柱为xxx C18 (250 mm*4.6 mm, 5 μm)，流动相A为甲醇，B为1% xxx溶液，流速为1.0 mL/min，柱温为25℃。
