2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,草酸,酒石酸,奎宁酸,苹果酸,抗坏血酸,柠檬酸,富马酸,琥珀酸标准品购于Sigma-Aldrch
All chemicals and solvent were HPLC. Standard: oxalic acid, tartaric acid, quinic acid, malic acid, ascorbic acid, citric acid, fumaric acid, succinic acid and were purchased from Sigma-Aldrch.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取1.0g左右样品;
2) 1.0g样品加入0.04 mol/L xxx溶液,冰浴下研磨成匀浆;
3) 超声提取20 min,冷冻条件下15000 r/min离心20 min,收集上清液,残留物重复提取;
4) 合并上清液,定容至10 mL,并经0.22 µm滤膜过滤,进样10 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Agilent高效液相色谱仪,配备二极管阵列检测器。
色谱柱为xxx ODS (250 mm*4.6 mm, 5 μm),流动相为0.04 mol/mL xxx-xxx缓冲液(pH=2.4),流速为0.5 mL/min,柱温为30℃。
