2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,原儿茶酸,儿茶素,绿原酸,咖啡酸,阿魏酸,芦丁标准品购于Sigma-Aldrch.
All chemicals and solvent were HPLC. Standard: protocatechuate, catechin, chlorogenic acid, caffeic acid, ferulic acid, rutin and were purchased from Sigma-Aldrch.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取10.0g左右的样品;
2) 10.0g样品加入60% xxx溶液,超声提取(功率250W,温度50℃)15 min;
3) 过滤,滤液用30% xxx溶液定容至25 mL,摇匀,用0.22 µm滤膜过滤,进样5 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Agilent 1100型高效液相色谱仪,配备二极管阵列检测器。
色谱柱为xxx-C18 (125 mm*4.6 mm, 5 μm),流动相A为xxx,B为0.01 mol/L xxx溶液,流速为0.8 mL/min,柱温为40℃。
