2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,乳糖,葡萄糖,半乳糖标准品购于xxx。
All chemicals and solvent were HPLC. Standard: lactose, glucose, galactose and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取5.0g左右的样品;
2) 5.0g样品加入25g水溶解,缓慢加入5 mL 10% xxx溶液;
3) 磁力搅拌30 min,冷却至室温,5000 r/min离心15min;
4) 取上清液用0.22 µm滤膜过滤,进样20 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Waters 2695型高效液相色谱仪,配备示差折光检测器。
色谱柱为xxx (300 mm*7.8 mm, 9 μm),流动相为5 mmol/L xxx溶液,流速为0.6 mL/min,柱温为60℃。
