2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,果糖,山梨糖醇,葡萄糖,蔗糖标准品购于xxx
All chemicals and solvent were HPLC. Standard: fructose, sorbitol, glucose, sucrose and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取2.0g左右样品;
2) 2.0g样品中加入20 mL xxx,研磨,4℃ 20000g离心15 min;
3) 取上清液,用0.45 µm滤膜过滤,进样10 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Agilent高效液相色谱仪,配备蒸发光散射检测器。
色谱柱为xxx NH2 (250 mm*4.6 mm, 5 μm),流动相为xxx-水(70:30, v/v),流速为1.0 mL/min,柱温为50℃。
